ABSTRACT

Objective:To investigate the expressions and significances of autophagy-related genes Beclin-1, LC3 and p62 in esophageal squamous cell carcinoma (ESCC).Methods:The clinical data of 112 patients with primary ESCC who underwent surgery at the 81st Group Army Hospital of Chinese PLA from January 2015 to December 2016 were retrospectively analyzed. Immunohistochemistry was used to examine the expressions of Beclin-1, p62 and LC3 proteins in 112 ESCC tissues and 31 adjacent normal esophageal mucosa tissues. Furthermore, the expressions of the above three autophagy-related markers in ESCC and the relationship between their expressions and the clinicopathological characteristics of patients were analyzed.Results:The positive expression rates of Beclin-1, LC3 and p62 in ESCC tissues were 32.14% (36/112), 37.50% (42/112) and 63.39% (71/112), The positive expression rates of Beclin-1, LC3 and p62 in adjacent normal esophageal mucosa tissues were 61.29% (19/31), 64.52% (20/31) and 32.26% (10/31), and the differences were statistically significant ( χ2 values ??were 8.715, 7.216 and 9.584, all P < 0.01). The positive expression rates of Beclin-1 and LC3 in ESCC were lower than those in adjacent normal esophageal mucosa tissues, and the positive expression rate of p62 in ESCC was higher than that in adjacentnormal esophageal mucosa tissues. In ESCC patients, the expression of Beclin-1 was related to histological grade, infiltration depth, TNM staging and lymph node metastasis (all P < 0.05); the expression of LC3 was related to infiltration depth and TNM staging (both P < 0.01); the expression of p62 was related to lymph node metastasis ( P < 0.01). In ESCC, the expression of LC3 was positively correlated with the expression of Beclin-1 ( r = 0.731, P = 0.001), and negatively correlated with the expression of p62 ( r = -0.215, P = 0.023). Conclusions:Autophagy plays a certain role in the occurrence and development of ESCC. Combined detection of autophagy-related genes Beclin-1, p62 and LC3 can assist clinical diagnosis and guide follow-up comprehensive treatment.

ABSTRACT

Objective To explore the best cut-off value of serum human epididymis protein 4(HE4)in the diagnosis of ovarian cancer,and analyze the clinical significance of combined detection of HE4,carbohydrate antigen 125 (CA125)and D-dimer for early screening and diagnosis of ovarian cancer.Methods A total of 49 patients with ovarian cancer,63 patients with benign tumor and 47 healthy women were collected.The content of CA125,HE4 and D-dimer were detected by chemical luminescence immunoassay and turbidimetric inhibition immunoassay,respectively.Results The new cut-off value of HE4 was 54.31 pmol/L.The serum levels of HE4,CA125 and D-dimer in the ovarian cancer group were all significant higher than those in the ovarian benign tumor group and the healthy group(P<0.05).No significant difference between the healthy group and benign disease group was found in CA125 and D-dimer content(P=0.293,0.359),while a significant difference in HE4 level(P<0.01).The sensitivity and specificity of combined detection of the HE4+CA125 increased to 91.8%,85.4%,which was higher than that of the diagnosis detection of the three indicators respectively.The sensitivity and specificity of combined detection of the HE4 +CA125 +D-dimer were 95.0%,76.5%,which was higher than the combined detection of the HE4+CA125.Conclusion The combined detection of CA125+HE4+D-dimer could improve the sensitivity for early diagnosis and screening of ovarian cancer.

ABSTRACT

[Abstract ] Objective Globular C1q receptor (gC1qR), a highly acidic receptor protein, is expressed in almost all mamma-lian cells in addition to exoerythrocytic, which can mediate a variety of biological responses.The study aimed to explore gC1qR expres-sion in cervical cancer and itd effects on the biological behaviors of human cervical cancer cells. Methods Retrospective analysis was made on 100 cervical tissue samples of patients in Nanjing Maternal and Child Health Hospital from August 2014 to April 2015, in-cluding 50 cervicitis tissues and 50 cervical cancer tissues.Immunohistochemical SP method was applied in the research of cervical cancer cell line C33a to detect the expression and the location of gC1qR in cervical tissues.Real-time PCR and Western blot analysis were respectively applied to detect the levels of gC1qR mRNA and gC1qR protein expression.Besides, the abilities of C33a cells mi-gration, invasion and apoptosis were respectively assessed by in vitro cell wound healing experiment, transwell assay and flow cytome-try. Results The expression of gC1qR gene was dramatically decreased in the group of cervical cancer tissues when compared with chronic cervictis group (2.18 ±0.37 vs 7.23 ±0.69, 0.27 ±0.09 vs 0.74 ±0.02, P cal cells by transmission electron microscope. Conclusion gC1qR expression might play an important role in inhibiting the invasion and migration of cervical cancer cells and inducing the apoptosis of cervical carcinoma cells, which provides new clues and potential targets for the treatment of cervical cancer in further research.

ABSTRACT

Objective To study the clinical effect of iodine-131 treatment of hyperthyroidism.Methods 92 hyperthyroidism patients were selected,who were randomly divided into the observation group and control group, with each 46 cases.The observation group was given radioactive iodine-131 treatment,the control group was given anti thyroid function of drug treatment.The efficacy and adverse drug reaction of the occurrence of long-term and short-term in the two groups were compared.Results In the observation group,the cure rate and total effective rate were respec-tively 93.48% and 100.00%,which were significantly higher than the control group for 63.04% and 84.78%(χ2 =12.55,7.57,all P <0.05).The incidence of adverse reaction in the observation group was 4.35%,which was significantly lower than 21.74% in the control group (χ2 =6.13,P <0.05).The recurrence and complication(hypothyroidism)cases in the observation group,were 1 cases and 8 cases,while the control group were 10 cases and 1 cases.The relapse rate of the observation group was significantly lower than that of the control group,and the incidence of compli-cations was significantly higher than the control group,the difference was statistically significant(χ2 =8.36,6.03,all P <0.05 ).Conclusion The iodine short-term efficacy of-131 in treatment of hyperthyroidism can significantly improve the treatment of the disease,and has high safety,but the complication rate is higher than that of anti thyroid drugs.

ABSTRACT

Objective To prepare the long‐term use and good stability of HBsAg feeble masculine serum ,and the application of controlling quality for HBsAg .Methods The HBsAg positive serum was diluted using calf serum phosphate buffer by grads ,multi‐ple to 1∶2 ,1∶4 ,1∶8 ,1∶16 ,1∶32;HBsAg feeble masculine serum was detected by double antibody sandwich method of enzyme‐linked immunosorbent assay (ELISA) using different reagent lot obtained from two companies for continuous six months .Results The serum ,s diluting multiple which the S/CO ,value between 1 to 3 was 1∶8 ,and the S/CO was 2 .42 ,the CV was 4 .72% .The serum of quality control possessed a good stability in different detection reagents ,the S/CO value range from 2 .41 to 2 .82 ,and the CV value was 4 .24% to 8 .68% .The CV value of HBsAg feeble masculine control serum with 6 months successive detection ranged from 8 .71% to 9 .86% (CV<10% ) .Conclusion The HBsAg feeble masculine serum possessed good stability and precision which is fit for masculine serum in HBsAg test .

ABSTRACT

Objective To investigate the application value of plasma D-dimer and coagulation indicators in the patients with o-varian cancer by detecting the routine coagulation indicators and plasma D-dimer level.Methods The plasma D-dimer and coagula-tion indicators levels were measured in 84 patients with ovarian cancer and 25 patients with benign ovarian tumor.The application value of preoperative plasma D-dimer level in monitoring the disease course of ovarian cancer was evaluated.Results The plasma levels of D-dimer and FBG in the ovarian cancer group were significantly higher than those in the ovarian benign tumor control group(P <0.01);moreover the plasma levels of D-dimer and FBG in the patients with the stage Ⅲ~Ⅳ ovarian cancer were signifi-cantly higher than those in the patients with the stageⅠ~Ⅱ ovarian cancer,the differences had statistical significance(P <0.05), which prompting that the coagulation and fibrinolysis dysfunction in the patients with ovarian cancer exhibited the aggravating trend with the disease condition progression.Conclusion D-dimer may reflect the excessive activation of fibrinolysis function in ovarian tumor,which prompting the survival period and prognosis in the patients with advanced ovarian cancer.The plasma D-dimer level has the important significance to early prevention,early diagnosis,early treatment and improve the prognosis in the thrombosis oc-currence in ovarian cancer.
